Long story short, I would like to paint over the back design on the old style Bicycle jumbo cards. Is this possible without it scratching or wearing off? Also, after painting they would have to be treated with roughing fluid.

I have an old DVD that talks about this. It is on the topic of making your own gimmicked cards.
The process is simple, lightly sand, paint or print the design (This DVD speaks of using an inkjet printer) and spray with Krylon spray to restore the "Air cushion" finish. I don't know anything about roughing it though. It might go right over the air cushion like with regular cards, but you'd have to trial and error that.
